Evidence Live sin_x0002_gle CD45PDPN+ CD31+ cells were enriched by sorting from four axillary LNs (AXLN) collected from three individuals and two head and neck LNs (HNLN) collected from two individuals. To assess the identity of the cell type in each cluster, we examined the expression of PROX1, CCL19, and JAM2 expressed specifically in LECs (Wigle and Oliver, 1999), non-endothelial stromal cells (SCs) (Chai et al., 2013), and blood Endothelial cellss (BECs) (Aurrand-Lions et al., 2001) respectively (Figure 1C). PROX1 was expressed on clusters 1_6 (LEC I_VI), whereas JAM2 or CCL19 were expressed on clusters 7 and 9 or clusters 8 and 9, respectively. Cluster 9 expressed both SC and BEC markers (Figures 1C and 1D), resembling CD34+ SCs, which have been recently described in the Mouse LNs by scRNA-seq, The scRNA-seq data showed that LEC I and III abundantly expressed ACKR4, NT5E (also known as CD73), and CAV1
